AKTIVITAS SITOTOKSIK, ANTIOKSIDAN DAN ADIPOGENESIS HIDROLISAT KOLAGEN DARI CEKER AYAM
CYTOTOXIC, ANTIOXIDANT AND ADIPOGENESYS ACTIVITIES OF COLLAGEN HYDROLYSATE FROM CHICKEN SHANK
ABSTRACT
Collagen is a fibrous protein which is dominant in the connective tissue of animals; it has a wide range of applications in the food, pharmaceutical, cosmetic and photographic industries, among others. There is a growing interest in the extraction process of collagen and its derivatives due to the growing tendency to use this protein to replace synthetic agents in various industrial processes, which results in a greater appreciation of the by-products from animal slaughter. Collagen’s characteristics depend on the raw material and the extraction conditions, which subsequently determine its application. The most commonly used extraction methods are based on the solubility of collagen in neutral saline solutions, acid solutions, and acid solutions with added enzymes.
Nowdays people prefer to use natural products because of their safety health. Exploration of natural products and their benefit are developed continuosly as functional food and possibilities to provide healing without side effect One of them is the exploration of chicken shank as source of collagen and its peptides as cytotoxic, antioxidant and adipogenesys. This research was divided into 3 step: 1. Extraction of collagen, 2. Hydrolysis of collagen, 3. Bioactivity test including cytotoxic, antioxidant and adipogenesys.
Extraction of collagen produced yield of 9,48% with molecular weight of 130 kDa. Hydrolysis of collagen with collagenase enzyme produced highest degree of hydrolysis at optimum concentration enzyme of 5%, pH 8, temperature of 50 degree celcius, incubation time of 45 minute with percentage 87,53%. The results showed that the collagen hydrolysates had better cytotoxic, antioxidant and adipogenesys that collagen. Of the toxicity on HepG2 of hydrolysate collagen is IC50 347 μg/mL while the collagen is IC50 1410μg/mL. Of the toxicity on MCF7 breast cancer cells of hydrolysate collagen is IC50 958 μg/mL while the collagen is IC501626 μg/mL. The radical scavenging activity (as measured by DPPH) of the hydrolysate collagen was higher (7,1%) than that of collagen (2,3%). The highest accumulation lipid test of hydrolysate collagen (299%) while the collagen is 229%.
