ISOLASI DAN UJI AKTIVITAS ANTIMIKROBA METABOLIT SEKUNDER KAPANG ENDOFIT DARI RANTING TANAMAN LEGUNDI (Vtex trifolia L.)

From previous studies it isrnknown that Legundi plant leaf extract (Vitex trifolia L.) have anrnantimicrobial activity. This study aims to isolate endophytic fungi fromrntwigs of Legundi plant and to test the antimicrobial potential of secondaryrnmetabolites from endophytic fungi isolates that were obtained. Endophyticrnfungi of twigs of Legundi plants are grown using direct planting methodrnusing a PDA. Results of insulation were obtained 6 isolates, then examinernmorphological observation of the endophytic fungi macroscopically (visual)rnand microscopic (slide culture), 4 single isolates were obtained. Each fourrnisolates then fermented using PDY media for 13 days. Supernatant fromrnfermentation then extracted gradually using n-hexane solvent, ethyl acetaternand n-butanol. Concentrated extract that were obtained from each solventrnexamined the antimicrobial activity using agar diffusion method againstrnStaphylococcus aureus and Escherichia coli and the yeast Candida albicansrnwith standard antibiotics chloramphenicol and nystatin. The result showedrnthat the concentrated extract of secondary metabolites of endophytic fungirnfrom n-butanol fraction capable of inhibiting all microbes, whereas thernfraction of n-hexane and ethyl acetate can only inhibit Staphylococcusrnaureus and Candida albicans, but can not inhibit Escherichia coli. From thisrnstudy it can be concluded that the secondary metabolites of endophytic fungirnfrom n-butanol fraction has antimicrobial activity against Gram-positive,rnGram-negative bacteria, and yeast, while the fraction of n-hexane and ethylrnacetate only has antimicrobial activity against Gram-positive bacteria andrnyeasts. rn rn(E) Literature: 38 (1983-2016)rn rn(F) Prof. Dr. Shirly Kumala, M.Biomed.,Apt. ; Dr. Priyo Wahyudi, M.Si.rn rn(G) 2016rn
